Under de föregående åren har neutrofila extracellulära fällor (NET) bevisats bidra till tillstånd av akut och kontinuerlig inflammatorisk sjukdom. De består av utdriven kromatin och pryds av neutrofila proteiner.
På grund av detta faktum har utvärderingen av DNA -komplex med myeloperoxidas ( MPO ) av ELISA blivit en vacker enhet för att mäta NET -bildning i in vitro- och in vivo -prover. När vi väl använde ett broadcast MPO -DNA ELISA -protokoll och inkluderade en isotyphantering för anti -MPO -beläggningsantikroppen, märkte vi överdriven analysspecificitet för in vitro -färdiga NET -prover, medan specificiteten för in vivo -plasmaprover var låg.
Förutom detekterade analysen inte in vitro -genererade MPO -DNA -komplex när de spetsades in i plasma. Slutligen upptäckte vi {att a} industriellt reagens avsett att dämma humana antimuskkroppar mot mus och multivalenta ämnen ökade detektionsfönstret mellan MPO- antikroppen och isotyphantering för extremt utspädd plasma.
Vi använde detta modifierade ELISA -protokoll för att undersöka MPO -DNA -komplex i mänskliga blodprov av akuta och kontinuerliga inflammatoriska situationer. Medan markörer för neutrofilaktivering och NET -bildning motsvarande MPO , elastas och citrullinerat histon H3 korrelerade avsevärt, märkte vi ingen korrelation med graderna av MPO -DNA -komplex.
På grund av detta faktum drar vi slutsatsen att ELISA -mätningar av MPO -DNA -komplex i mänsklig plasma är extremt tveksamma när det gäller specificitet av NET -detektion. I allmänhet borde plasmaanalyser av ELISA extra ofta omfamna isotypkontroller för antikroppar för att avslöja målspecificitet.
JAK3 och PI3K förmedlar de undertryckande resultaten av interferon tau på neutrofila extracellulära fällor under hela peri-implantationsintervallet
Interferon tau (IFNτ) är det huvudsakliga moderns tecken på att vara gravid hos idisslare och modulerar förmågan hos olika immunceller, tillsammans med neutrofiler. Neutrofila extracellulära fällor (NET) är en av många viktiga försvarsmekanismer för neutrofiler.
Vid denna undersökning märkte vi ökad (p <0,01) ex-vivo NET-extrudering av blodneutrofiler från dag 16-18, som satte upp syntetisk insemination (AI) hos icke-inseminerade och inseminerade icke-dräktiga kor i jämförelse med dräktiga kor. In vitro -undersökning bekräftade dessutom att IFNτ hämmar bildandet av NET i dos- och tidsberoende metod.
Botten (p <0,01) NET-bildningen och det bästa (p <0,01) mRNA-uttrycket (RT-PCR) för IFNτ-stimulerade gener (ISG15, OAS1, MX1) har märkts när neutrofil inkuberades med 9 ng/ml IFNτ i 3,5 timmar .
Signalkaskader som förmedlar IFNτ -försämring av NETs -bildning har erkänts med hjälp av hämmare av JAK2, JAK3, p38, PI3K/Akt och MAPK/Erk. IFNτ sänkte (p <0,01) mRNA-uttrycket (RT-PCR) och fokus ( ELISA ) för gener och proteiner som medierar bildandet av NET i blodneutrofiler tillsammans med histoner (H1, H2), neutrofilt elastas (NE) och myeloperoxidas ( MPO ) .
Ändå har resultaten av IFNτ på dessa gener och proteiner utrotats i närvaro av JAK3- eller PI3K -hämmare. Immunocytokemi undersöker ytterligare bekräftat robust MPO -tecken i närvaro av JAK3- eller PI3K -hämmare jämfört med optimistisk hantering (PC, IFNτ ensam). Resultaten pekar på att IFNτ försämrar bildandet av NET med JAK3 och PI3K och därmed viktigt för lönsam implantation och institution för att vara dräktig hos kor.
Överdriven omgivningstemperatur förvärrar experimentella autoimmuna uveit -tecken
Huruvida omgivningstemperaturen påverkar immunsvaret som resulterar i uveit är okänt. Vi undersökte således om omgivningstemperaturen påverkar tecknen på experimentell autoimmun uveit (EAU) hos möss eller inte och undersökte genomförbara mekanismer. C57BL/6-möss har lagrats vid en standard (22 ° C) eller hög temperatur (30 ° C) i två veckor och har sedan immuniserats med humant interfotoceptor-retinoidbindande protein (IRBP651-670) peptid för att inducera EAU.
Histologiska justeringar har övervakats för att bedöma svårighetsgraden av uveit. Frekvensen för Thl -celler och Th17 -celler mättes med cirkulerande cytometri (FCM). Uttrycket av IFN-y och IL-17A mRNA mättes med realtid qPCR. Tekniken för neutrofila extracellulära fällor (NET) kvantifierades genom enzymkopplad immunosorbentanalys ( ELISA ).
Differentialmetaboliter inom plasman hos mössen som lagrats inom de ovannämnda två omgivningstemperaturerna har mätts med hjälp av ultrahögpresterande vätskekromatografi trippel kvadrupol masspektrometri quadrupol tid för flygmasspektrometri (UHPLC-QQQ/MS). De differentialmetaboliter som känns igen har använts för att bedöma deras resultat på differentiering av Th1- och Th17 -celler och teknik för NET in vitro.
Resultaten bekräftade att EAU -möss lagrade vid överdriven temperatur skickade en extra extrem histopatologisk manifestation av uveit än möss som lagrats vid en standardtemperatur. En avsevärt förhöjd frekvens av Thl- och Th17-celler i anslutning till ett uppreglerat uttryck av IFN-y och IL-17A-mRNA märktes i mjältlymfocyterna och näthinnorna hos EAU-möss vid överdriven temperatur.
Uttrycket av NET, vilket framgår av myeloperoxidas ( MPO ) och neutrofilt elastas (NE), var avsevärt förhöjt i serum och supernatanter av neutrofiler från EAU -möss lagrade vid för hög temperatur i jämförelse med den konventionella temperaturgruppen.
Metaboliterna i plasma från EAU-möss, fumarsyra och ravsyra, har ökat markant inom gruppen för höga temperaturer och kommer att inducera tekniken för NET via NADPH-oxidasberoende väg, men påverkade inte frekvensen av Th1 och Th17 -celler.
Våra resultat rekommenderar att en förhöjd omgivningstemperatur är en hotfråga vid uveit. Det är relaterat till induktion av Th1- och Th17-celler utöver tekniken för NET som kan förmedlas av den NADPH-oxidasberoende vägen.
Simvastatin dämpar akut lungskada genom aktivering av A2B -adenosinreceptor
För att upptäcka skyddsmekanismen för simvastatin vid akut lungskada (ALI) användes den lipopolysackarid (LPS) inducerade (5 mg/kg) ALI -råttdockan för att titta på resultaten av simvastatin. I synnerhet sänkte simvastatin graderna av A2BAR, CFTR och claudin18 men uppreglerade claudin4 i lungvävnader. Dessutom motverkade terapi med PSB1115, en antagonist av A2BAR, den skyddande effekten av simvastatin vid ALI. Vår undersökning visar att simvastatin har en skyddande effekt i motsats till LPS-inducerad ALI genom att aktivera A2BAR och måste utnyttjas som ett nytt terapeutiskt mål för behandling av ALI.
