Akut lungskada (ALI) är en akut sjukdom som saknar effektiva behandlingar, vilket leder till jämförelsevis överdriven sjuklighet och dödlighet. Den patologiska mekanismen för ALI rapporteras ha samband med extrem irritation och aktiverad oxidativ stress. Nuvarande undersöker mål för att analysera skyddsresultaten av DPP-Four-hämmaren Trelagliptin i motsats till lipopolysackarid (LPS) -inducerad ALI och den bakomliggande mekanismen. LPS användes för att inducera ALI -möss mode.
Den patologiska situationen för ALI -möss utvärderades med användning av MPO -träningsanalys, detektering av lungfuktighet till torrvikt och HE -färgning på lungvävnaderna. Lungoperationen bedömdes med hjälp av en spirometer. Det oxidativa stresssteget i lungvävnaderna kontrollerades med MDA -mätning och GPx -detektion med användning av industriella kit.
Leukocyt- och neutrofiltalet hade bestämts med hjälp av en hemocytometer och proteinets fulla fokus inom BALF detekterades med användning av en bikinchoninsyrateknik. Uttrycksintervallen för TNF-a, IL-6 och CXCL2 i lungvävnaderna hade utvärderats med användning av qRT-PCR och ELISA .
Western blot-utvärdering användes för att ta reda på uttrycksintervallen för TLR4 och p-NF-KB p65. LPS-inducerad förhöjd MPO- övning, lungfuktighet till torrviktsförhållande, luftvägsresistens (RAW), alla typer av leukocyter och neutrofiler, tillverkning av inflammatoriska komponenter, minskad lungdynamisk efterlevnad (Cdyn) och maximal expiratorisk cirkulation (PEF) och ett förvärrat histopatologiskt tillstånd (jämförbart med störd alveolär konstruktion, viktigt pulmonärt interstitiellt ödem och ett stort antal röda blodceller och inflammatoriska celler i alveolär kavitet) hade avsevärt reverserats genom administrering av Trelagliptin.
TLR4/NF-KB signaleringsvägen aktiverades och oxidativ stress inducerades genom stimulering med LPS; dock hade varje resultat undertryckts genom administrering av Trelagliptin. Trelagliptin kan lindra LPS-inducerad irritation och oxidativ stress hos möss med akuta lungskador.
Diammoniumglycyrrhizinat lipidligand förbättrar lipopolysackaridinducerad akut lungskada genom modulering av vaskulär endotelbarriär
Avsikten med den aktuella undersökningen var att analysera konsekvenserna av diammonium glycyrrhizinate lipid ligand (DGLL) terapi på akut lungskada (ALI) och lungödem inducerat av lipopolysackarid (LPS) hos Sprague-Dawley råttor. Råttor erhöll oralt 30, 60 och 120 mg/kg DGLL. Efter 1 timma fastställdes ALI -mannequin från råtta med LPS (10 mg/kg) intraperitoneal injektion. Efter 6 timmar utvärderades lungskada med användning av hematoxylin- och eosinfärgningsmetoder.
Lungödem utvärderades med användning av lung-våt-torr-viktförhållande, proteinkoncentrationer i bronchoalveolär sköljvätska (BALF) och Evans blue (EB) extravasation i lungvävnad. Uttrycksområdena för tumörnekrosfråga (TNF) -α och interleukin (IL) -1p i lungvävnader hade mätts med ELISA . Myeloperoxidas ( MPO ) uttrycksintervall hade detekterats genom immunhistokemisk färgning.
Western blotting användes för att mäta expressionsstadiet modifieringar av intercellulär vidhäftningsmolekyl (ICAM) -1, förutom vidhäftande och tighta övergångsproteiner, tillsammans med vaskulärt endotel (VE) -cadherin, zonula occludens (ZO) -1, occludin och junctional vidhäftningsmolekyl (JAM) -1 som hade varit relaterad till lungirritation och mikrovaskulär permeabilitet. DGLL-terapi lindrade avsevärt ALI inducerat av LPS, vilket demonstrerades genom rabatt på MPO- positiva celler och uttrycksintervall för TNF-α, IL-1β och ICAM-1 i lungvävnader från råtta.
Förutom att DGLL upphävde LPS-inducerat lungödem, minskade proteinfokus i BALF och sänkte EB-extravasation. DGLL reverserade dessutom det sänkta uttrycket av VE-cadherin och tight junction-proteiner, tillsammans med ZO-1, occludin och JAM-1 i lungvävnaderna som orsakas av LPS.
Sammanfattningsvis avslöjar DGLL en skyddande effekt på LPS-inducerad råtta ALI, vilket är relaterat till hämning av inflammatorisk cellinfiltration och mikrovaskulär barriärstörning. De nuvarande resultaten presenterar en teoretisk grund för DGLL -apparaten för potentiell vetenskaplig terapi av ALI.
Madecassoside skyddar mot LPS-inducerad akut lungskada genom att hämma TLR4/NF-κB-aktivering och blod-luftbarriärpermeabilitet
Madecassoside (MA), en viktig ingrediens i Centella asiatica , har rapporterats uppvisa ett brett spektrum av bioaktiviteter, tillsammans med antipulmonell fibros och antiinflammatoriska resultat. Just här syftade vi till att belysa skyddsresultaten och underliggande mekanismer för MA på LPS-inducerad akut lungskada (ALI). Mössen hade hanterats med MA i en vecka, varefter intratrakealt LPS erhölls för att fastställa ALI -skyltdockan.
Därefter utvärderade vi de patologiska modifieringarna genom hematoxylin- och eosinfärgning och mätte graderna av proinflammatoriska cytokiner och myeloperoxidas ( MPO ) med ELISA , transkriptionsstadiet av tight junction-proteiner med qRT-PCR, förutom uttrycket av Toll-liknande receptor4/Nuclear utfärda kappa-B (TLR4/NF-κB) -väg med Western blot. Resultaten bekräftade att MA avsevärt hämmade LPS-inducerade patologiska skador, lungödem, MPO och proinflammatoriska cytokiner.
Dessutom reparerade MA tydligt alveolär epitelintegritet som visades genom minskad utsöndring av fullständiga proteiner inom BALF och förbättrat mRNA-uttryck för täta korsningar som Occludin och zonula occludens-1 (ZO-1) utvärderas till LPS. Ytterligare analys bekräftade att LPS-stimulering aktiverade TLR4/NF-κB-signalvägen och aktiveringen hämmades av MA.
Sammanfattningsvis indikerade denna information att MA hade skyddande resultat i motsats till LPS-inducerad ALI. De terapeutiska mekanismerna kan också relateras till minskning av alveolär epitelpermeabilitet och inflammatoriskt svar genom att undertrycka aktiveringen av TLR4/NF-KB-vägen.
Bindning av laktoferrin till golvet i lågdensitetslipoproteiner modifierade av myeloperoxidas förhindrar intracellulär ldl-kolesterolackumulering av humana blodmonocyter
Myeloperoxidas ( MPO ) är exklusivt heminnehållande peroxidas som kan katalysera bildning av hypoklorsyra (HOCl). Robust samspel av MPO med lågdensitetslipoproteiner (LDL) främjar proaterogen modifiering av LDL med HOCl. Det så kallade MPO- modifierade LDL (Mox-LDL) ackumuleras i makrofager med bildning av skumceller, vilket är det patognomiska symptomet på åderförkalkning. En lovande metod för profylax och botemedel mot åderförkalkning försöker hitta behandlingar som stoppar modifiering eller ackumulering av LDL i makrofager.
Laktoferrin (LF) har ett antal mjukvarufaktorer för viktproblempatogenes. Vi siktade på att kontrollera LF-bindning till Mox-LDL och deras ackumulering i monocyter omarbetade till makrofager. Genom att använda golvplasmonresonans och ELISA- metoder märkte vi inget LF-samspel med intakt LDL, medan Mox-LDL starkt interagerade med LF.
