Diabetic Bone Marrow Cell Injection Accelerated Acute Pancreatitis Progression
Akut pankreatit (AP) är sannolikt en av huvudorsakerna till sjukhusinläggning, varav 20% kan utvecklas till den extrema sorten med djupgående acinarcellnekros. Medicinsk forskning har rapporterat att diabetes är en opartisk riskfråga om förekomsten av AP och är relaterad till ökad svårighetsgrad än ämnen utan diabetes. Men hur diabetes deltar i AP -utvecklingen beskrivs inte effektivt.
För att analysera denna fråga hade vildtyp (wt) och diabetiska db/db-möss vid 16 veckors ålder använts inom forskningen. AP inducerades i viktmottagare med 10 injektioner av 50  μg /kg caerulein med ett intervall på 1 timme. En timme efter den sista caerulein -injektionen hade benmärgsceller (BMC) remoterade från wt- och db/db -möss injicerats intraperitonealt i mottagarna (1 x 107 celler/mottagare).
Mottagarna utan BMC -injektion fungerade som kontroller. 13 timmar efter BMC-injektion ökade serumlipasövningen 1,8- och 1,3-faldigt hos möss som förvärvade db/db BMC, i motsats till dessa utan injektion respektive BMC-injektion ( p  ≤ 0,02 för varje).
Genom H & E -färgning var den allmänna allvarlighetsgraden 14,7 för ingen cellinjektion och 16,6 för BMC -injektion i vikt och förhöjd till 22,6 för db/db BMC -injektion ( p  ≤ 0,002 för varje). Specifikt utvecklade möss med db/db BMC -injektion extra acinarcellnekros och vakuolisering än motsatta lag ( p  ≤ 0,03 för varje).
När sektioner hade färgats med en antikropp i motsats till myeloperoxidas ( MPO ) var densiteten hos  MPO + -celler vid pankreatit 1,9- och 1,6-faldig ökad än vikt BMC och inga BMC-injektionsteam, individuellt ( p  ≤ 0,02 för varje).
Kvantifierat med  ELISA , db/db BMC producerade extra IL-6, GM-CSF och IL-10 i motsats till vikt BMC ( p ≤ 0,04 för alla). Sammanfattningsvis producerade BMC för db/db -möss extra inflammatoriska cytokiner. Som svar på acinar cellskada förvärrade diabetisk BMC irritationskaskaden och acinar cellskador, vilket resulterade i utvecklingen av akut pankreatit.

Corynoline skyddar lipopolysackaridinducerad mastit genom att reglera AKT/GSK3β/Nrf2 signalväg

Irritation har identifierats vara oroande vid patogenesen av mastit. Och antiinflammatoriskt medel föreslås vara en uppnåelig miljövänlig terapeutisk teknik för mastit. Corynoline, en bioaktiv förening extraherad från Corydalis bungeana Turcz., Har rapporterats ha antiinflammatorisk effekt.
Huruvida korynolin har en skyddande effekt i motstånd mot mastit förblir dock oklart. Avsikten med denna forskning var att bedöma den skyddande effekten av korynolin på LPS-inducerad mastit hos möss. Inflammatorisk cytokintillverkning mättes med  ELISA . Proteinerna i signalvägar hade detekterats genom western blot -utvärdering.
Resultaten bekräftade att botemedel mot korynolin vid doserna 15, 30 och 60 mg/kg avsevärt försvagade LPS-inducerad patologisk skada på bröstvävnader. Korynolin förbättrade dessutom LPS-inducerad  MPO- träning, MDA-innehållsmaterial och inflammatorisk cytokin TNF-a och IL-1β-tillverkning i bröstvävnader.
LPS-inducerad NF-KB-aktivering inhiberades av korynolin. Dessutom bekräftade våra resultat korynolin avsevärt uttrycket av Nrf2 och fosforyleringsintervallet för AKT och GSK3β. Sammanfattningsvis indikerade våra resultat att korynolin skyddade mot LPS-inducerad mastit genom att reglera AKT/GSK3β/Nrf2 signalväg, vilket senare ledde till inhibering av NF-KB och inflammatoriskt svar.

Taohong Siwu -avkok reglerar cellnekros och neuroinflammation inom råttcentret Cerebral Artery Occlusion Mannequin

Cellnekros och neuroinflammation spelar en nödvändig funktion för att tänka på skador som orsakas av ischemisk stroke. Tidigare forskning har rapporterat att Taohong Siwu-avkok (THSWD) kan skala ner koronar hjärtmuskelnekros och har antiinflammatoriska egenskaper. På denna forskning undersökte vi konsekvenserna av THSWD på cellnekros och neuroinflammation i en råttdocka med center cerebral arterie occlusion (MCAO).
Trettiosex hanråttor Sprague-Dawley (SD) hade slumpmässigt delats in i tre lag med 12 råttor i varje grupp. De hade varit skenoperationsgruppen, MCAO -mannequingruppen och MCAO + THSWD -gruppen. Vi använde  ELISA för  att ta reda på graderna av TNF-α, Mcp-1 och IL-1β inflammatoriska element i råttserum, qRT-PCR för att detektera uttrycket av TNF-α, Mcp-1 och IL-1β mRNA i råtta , och immunhistokemi för att upptäcka variationen av mikroglia och neutrofiler i råtta. qRT-PCR och Western blot hade använts för att detektera mRNA- och proteinuttrycksintervall för IBA- 1- och  MPO-  inflammatoriska element och TNF-a/RIP1/RIP3/MLKL-vägen inom råttans sinne och proteinuttrycksintervall.
I motsats till skenoperationsgruppen hade uttrycket av MCP-1, IL-1β, IBA-1 och  MPO  inflammatoriska element och TNF-a/RIP1/RIP3/MLKL-vägen väsentligt uppreglerats inom MCAO-gruppen. I motsats till MCAO-gruppen hade uttrycken av MCP-1, IL-1β, IBA-1 och  MPO-  inflammatoriska element och TNF-α/RIP1/RIP3/MLKL-vägen avsevärt nedreglerats inom MCAO + THSWD-gruppen.
THSWD kan skala tillbaka uttrycksintervallen för MCP-1, IL-1β, IBA-1 och  MPO  inflammatoriska element utöver TNF-α/RIP1/RIP3/MLKL-vägen. Under tiden kan det eventuellt skala ner nekros och irritation av hjärnceller efter cerebral ischemi, för att skydda hjärnvävnaden hos råttor.

Dihydrotanshinon dämpar LPS-inducerad akut lungskada hos möss genom uppreglering av LXRα

Dihydrotanshinon (DIH) är ett extrakt av Salvia miltiorrhiza Bunge. Det har rapporterats att DIH kan reglera NF-KB-signalvägen. Avsikten med denna forskning var att undersöka huruvida DIH kan skydda möss från lipopolysackarid (LPS) -inducerad akut lungskada (ALI) hos möss. I denna forskning hade sextio möss slumpmässigt delats in i 5 lag, en grupp som ren ledningsgrupp, den andra gruppen som LPS-ledningsgrupp och de tre sista lagen hade förinjicerats med helt olika doser av DIH, varefter inandade LPS för experimentell jämförbarhet.
Efter 12 timmars behandling med LPS hade våt-torr-förhållandet, histopatologiska justeringar och myeloperoxidas ( MPO ) övning av lungorna mätts. Förutom hade  ELISA-  kit använts för att mäta graderna av TNF-α och IL-1β inflammatoriska cytokiner i bronkoalveolära sköljvätskor (BALF), och western blot-utvärdering användes för att mäta utövandet av NF-κB-signalväg.
Resultaten visade att DIH framgångsrikt skulle kunna minska lungödem,  MPO  -träning och förbättra lunghistopatologiska justeringar. Dessutom undertryckte DIH graderna av inflammatoriska cytokiner i BALF, liknande TNF-α och IL-1β.
Förutom att DIH ytterligare kan nedreglera utövandet av NF-κB-signalvägen. Vi upptäckte dessutom att DIH dosberoende höjde uttrycket av LXRa. Likaså kan DIH hämma LPS-inducerad IL-Eight-tillverkning och NF-KB-aktivering i A549-celler.
Och de hämmande resultaten hade reverserats av LXRa -hämmare geranylgeranylpyrofosfat (GGPP). På grund av detta faktum spekulerar vi att DIH reglerar LPS-inducerat ALI hos möss genom att öka LXRa-uttryck, vilket därefter hämmar NF-KB-signalvägen.

Protectin D1 skyddar i motsats till lipopolysackaridinducerad akut lungskada genom hämning av neutrofilinfiltration och bildandet av neutrofila extracellulära fällor i lungvävnad

Protectin D1 (PD1), en DHA-härledd lipidförmedlare, har inte för länge sedan visat sig ha antiinflammatoriska och pro-upplösande egenskaper. Hittills är lite förstått om PD1: s inverkan på lipopolysackarid (LPS) -inducerad akut lungskada (ALI) hos möss.
Avsikten med den aktuella forskningen var att undersöka de terapeutiska resultaten av PD1 på LPS-inducerad ALI och dess potentiella rörelsemekanismer. ALI inducerades genom en intraperitoneal injektion av LPS, platsen PD1 (2 ng/mus) administrerades intravenöst 30 minuter efter LPS -problem. Möss hade offrats 24 timmar efter modellering.
Lunghistopatologiska justeringar hade utvärderats med hjälp av hematoxylin och eosinfärgning och myeloperoxidas ( MPO ) träning undersöktes med hjälp av immunhistokemi. Tumörnekros-α och interleukin-6-intervall inom bronkoalveolär sköljvätska (BALF) och serum hade mätts med  ELISA .
För att detektera neutrofila extracellulära fällor producerade av infiltrerade neutrofiler i lungvävnaden utfördes immunofluorescensfärgning med användning av anti- MPO-  och anti-histon H3-antikroppar. Resultaten indikerade att PD1 avsevärt dämpade histologisk skada och neutrofilinfiltration i lungvävnad, minskade lungfukt/torrviktsförhållandet, proteinfokus och proinflammatoriska cytokinintervall i BALF och minskade proinflammatoriska cytokinintervall i serum.
